What is Doxycycline?

Doxycycline is a semisynthetic bacteriostatic antibiotic that belongs to a group of tetracycline antibiotics. Doxycycline posesses a broad spectrum of antibacterial activity. It works by inhibiting protein synthesis in the microbial cell at the ribosomal level. Doxycycline acton intracellularly located pathogens (mycoplasma, chlamydia, rickettsia). What is Doxycycline action mechanism and spectrum of activity?

Doxycycline is a semisynthetic broad-spectrum antibiotic of tetracycline group. Doxycycline posesses bacteriostatic action by penetrating into the bacterial cell. Action mechanism is associated with Doxycycline interferes with the connection of transport aminoacyl-RNA to 30S ribosomal subunit and thus inhibits protein synthesis in the microbial cell at the ribosomal level. Aerobic bacteria: Escherichia coli, Shigella spp., Salmonella spp., Yersinia spp., Enterobacter spp., Entamoeba histolytica, Klebsiella spp., Bordetella pertussis, Haemophilus influenzae, Pasteurella spp., Bacteroides spp. Doxycycline is also active against Rickettsia spp., Treponema spp. , Mycoplasma spp. and Chlamydia spp. Haemophilus influenzae (91-96%) and intracellular pathogens (Chlamydia, mycoplasma, rickettsia) are the most susceptive to doxycycline.

Doxycycline is active against most dangerous infectious diseases: plague, tularemia, anthrax germs, Legionella, Brucella, cholera, glanders germs, pathogens ornithosis, psittacosis, trachoma, venereal granuloma. Which are Doxycycline pharmakokinetic characteristics?

After oral administration Doxycycline is almost completely absorbed from the gastrointestinal tract into the blood circulation. It has a high degree of lipid solubility and meal has no significant effect on the absorption degree. The maximum concentration in plasma usually is determined in 2 hours after administration. Doxycycline Binds to plasma proteins by 80-95%. Doxycycline is widely distributed in tissues and body fluids. • Infections of skin and soft tissue (acne, cellulitis, abscesses, furunculosis, whitlow, infected burns and wounds)

Doxycycline should not be used in the following conditions:

Doxycycline is contraindicated for use during pregnancy. It can penetrate through the placental barrier and cause permanent discoloration of teeth, enamel hypoplasia, inhibition of bone growth of the fetus. The medication exerts mutagenic effects. Doxycycline is determined in breast milk in quantities that make up 30-40% from its level in serum. Therefore the use of the medication during breastfeeding should be avoided.

How Doxicip 100 can interact with other medicines?

You should not Doxycycline simultaneously with antacids containing aluminum salts, calcium and magnesium (Phosphaluger, Maalox, Rennie et al), iron supplementation (iron salts, ferric hydroxide), sodium bicarbonate, magnesium containing laxatives (magnesium sulfate) Absorption of doxycycline is reduced in simultaneous use wuth cholestyramine and colestipol. Barbiturates, carbamazepine, phenytoin, rifampin, and other inductors of microsomal oxidation accelerates the metabolism of doxycycline and reduce the period of its half-life due to the acceleration of metabolism in the liver. Doxycycline reduces prothrombin index and potentiates the effect of indirect anticoagulants (warfarin, fenilin, etc.).

Doxycycline reduces the effectiveness of bactericidal antibiotics that act on bacterial cellular wall. Doxycycline hydrochloride reduces the effect of penicillins (benzylpenicillin, benzathine benzylpenicillin, oxacillin, amoxicillin, amoxicillin and clavulanate), cephalosporins (cefazolin, cefuroxime, cefotaxime, cefepime, ceftriaxone, ceftazidime, cefoperazone, cephalexin, cefixime, cefaclor).

Doxycycline reduces the effectiveness of oral birth control pills and increases the risk of bleedings. Simultaneous application with retinol leads to increased intracranial pressure. Doxycycline enhances the action of antidiabetic drugs: sulfonylurea derivatives (glibenclamide, glipizide, glimepirid). Doxycycline enhances the effects of nephrotoxic drugs.

Doxycycline hydrochloride increases the toxicity of methotrexate and cyclosporine A. The medication can also increase the risk of gastrointestinal side effects when used in combination with theophylline.
