This is beneficial for patients with insufficient blood flow to the penis. 

Physiological mechanism of erection consists in release of NO (nitrogen oxide) in response to sexual stimulation. The released NO interacts with the enzyme guanylate cycles and increases the amount of cyclic GMP. Cyclic GMP binds to specific proteins and causes them to relax. This leads to the relaxation of the smooth muscles of the penis and erection occurs.

When Vardenafil is prescribed?

Vardenafil drug is prescribed only for patients with erectile dysfunction. Erectile dysfunction is manifested in inability to achieve erection enough for the sexual act. Vardenafil hci acts naturally meaning that it is ineffective without sexual arousal. Vardenafil should be used only in case of sexual problems like erectile dysfunction and should not be used in healthy patients to improve their erection. Which is Vardenafil dosage?

The drug is taken orally with or without food. Initial recommended dose is 10 mg 25-60 minutes before sexual intercourse. The medication can be taken 4-5 hours to 25 minutes before sexual activity. The maximum frequency of intakes - 1 time daily. The medication is ineffective without sexual stimulation.

Depending on the efficacy and tolerability the dose may be increased to 20 mg or decreased to 5 mg  per day. The maximum daily dose is 20 mg.

·         Simultaneous use of nitrates and nitrogen oxide donors
·         Simultaneous use of HIV protease inhibitors
·         Women
·         Children under 18 years of age
 
Application during pregnancy and breastfeeding

Vardenafil is contraindicated for women and therefore the medication should not be taken during pregnancy and breastfeeding

Vardenafil side effects

Like other medications Vardenafil may cause side effects. However, it does not mean that all people taking Vardenafil tablets will experience any of these side effects. Possible Vardenafil side effects may include:

Central nervous system and peripheral nervous system: ≥ 10% - headache; ≥ 1% - dizziness; ≥ 0.1% - <1% - drowsiness; ≥ 0.01% - <0.1% - anxiety, fainting.
Cardiovascular system: ≥ 10% - hot flashes (sudden intermittent facial flushing, sensation of heat); ≥ 0.1% - <1% - increase in blood pressure, lower blood pressure, orthostatic hypotension; ≥ 0.01% - <0.1% - angina, ischemia infarction.
Digestive system: ≥ 1% - <10% - dyspepsia, nausea; ≥ 0.1% - <1% - change in liver function tests (increased ALT, AST, GGTP).
Respiratory system: ≥ 1% - <10% - congestive hyperemia of the nasal mucosa (mucosal edema, rhinitis, rhinorrhea); ≥ 0.1% - <1% - dyspnea, epistaxis; ≥ 0.01% - <0.1% - swelling of the larynx
Vision organs: ≥ 0.1% - <1% - increased tearing, blurred vision (luminance of view); ≥ 0.01% - <0.1% - increased intraocular pressure.
Dermatological reactions: ≥ 0.1% - <1% - face edema, photosensitivity.
Musculoskeletal system: ≥ 0.1% - <1% - myalgia, back pain, increased CPK; ≥ 0.01% - <0.1% - increase in muscle tone.
Reproductive system:> 0.01% - <0.1% - elongation erection or painful erections, priapism.
Other: ≥ 0.01% - <0.1% - hypersensitivity reactions.

There are post-marketing reports of cases of anterior ischemic optic neuropathy leading to impairment, including the persistent loss of vision, related in time with receiving PDE5 inhibitors, including and Vardenafil in patients, many of whom have concomitant risk factors for developing this condition, such as an anatomical defect of the optic nerve, being over 50 years of age, diabetes mellitus, hypertension, coronary artery disease, hyperlipidemia and smoking. It is unknown whether the development of anterior ischemic optic neuropathy is directly related to use of PDE5 inhibitors.

Can Vardenafil interact with other drugs?

Yes. Vardenafil should not be taken in certain medications. With other drugs Vardenafil dose should not exceed 5mg once daily. Vardenafil can interact with the following medicines:

Inhibitors of liver's isoenzymes such as CYP3A4, CYP3A5 è CYP2C9 of the system cytochrome P450 such as cimetidine, ketoconazole, itraconazole, indinavir, ritonavir, erythromycin. These medicines lowers Vardenafil clearance

Nitrates such as nitroglycerin and other nitrogen oxide donors when used with PDE5 inhibitors may cause extremely low blood pressure

Alpha blockers when used with Vardenafil may cause extremely low blood pressure
