What is Synthroid? 

Synthroid (levothyroxine sodium) is a synthetic levogyrate isomer of thyroxine (natural thyroid gland hormone). After a partial conversion to triiodothyronine in the liver and kidneys it enters the body's cells where regulates development and growth of tissues and activate metabolic processes. In low doses (Synthroid 25mcg), levothyroxine exerts anabolic effects on protein, lipid and carbohydrate metabolism. In medium doses, levothyroxine (Synthroid 100mcg) stimulates the growth of tisse, increases demand of tissues in oxygen,  increases the functional activity of cardiovascular system and central nervous system. 

In high doses, Synthroid (200mcg) inhibits the production of thyrotropin-releasing hormone, hypothalamus and pituitary thyroid stimulating hormone.
Therapeutic effect is usually observed after 7-12 days. Clinical effect of hypothyroidism treatment is manifested in 3-5 days. Diffuse goiter is reduced or disappears within 3-6 months. 

Which is Synthroid pharmacokinetics?  

After oral administration levothyroxine sodium is absorbed  in the upper part small intestine. Absorption rate is nearly 80% of the administered dose. Meal reduces the absorbtion of levothyroxine.
Cmax in serum is reached approximately in 5-6 h after administration. 
Levothyroxine binds to plasma proteins by 99% (thyroxine-dependent globulin, thyroxine-binding prealbumin and albumin). Nearly 80% of Levothyroxine is converted in  triiodothyronine (T3 thyroid  hormone) in the liver and kidney.  Thyroid hormones are metabolized primarily in liver, kidney, brain and muscle. Metabolites are excreted in the urine and bile. Half-life period is 6-7 days.

Which conditions are treated with Synthroid? 

Synthroid is used to to treat conditions associated with insufficiency of thyroid hormones. 

These conditions include:       

  • Hypothyroidism        
  • Euthyroid goiter         
  • As a replacement therapy and prophylaxis if goiter prophylaxis after thyroid gland resection
  • Thyroid cancer (after operative treatment)          
  • In diagnostics of thyroid suppression test      

Which is Synthroid dose and it is taken? 

The dose of the medication depends on the particular condition treated. The medication comes in the form of tablets containing Synthroid 25mcg, Synthoid 50mcg and Synthroid 100mcg. It is usually taken once a day in the morning on an empty stomach.   

When Synthroid tablets are contraindicated? 

Synthroid contraindications include:       

  • Thyrotoxicosis        
  • Hypopituitarism (pituitary insufficiency)      
  • Untreated insufficiency of adrenal cortex     
  • Acute heart attack (myocardial infarction)         
  • Acute pancarditis       
  • Acute myocarditits        
  • Known cases of hypersensitivity to levothyroxine sodium or other non-active components of the medication     

Synthroid should be used with caution in:         

  • Atherosclerosis        
  • Stenocardia         
  • Myocardial infarction in anamnesis         
  • Hypertension          
  • Arrhythmia        
  • Diabetes          
  • Long-term hypothyroidism         
  • Malabsorption syndrome       

Which Synthroid side effects are possible during the treatment? 

In most cases Synthroid does not any side effects because its ingredient is analogue of natural thyroid hormone. In high doses Synthroid (100mcg) or Synthroid 200mcg may cause the following side effect associated with hyperthyroidism: ·         

  • Increased sweating          
  • Fever         
  • Anxiety         
  • Insomnia         
  • High blood pressure          
  • Mood changes         
  • Increased heart rate       
  • Diarrhea    
  • Hair loss       
  • Flushing       
  • Menstrual problems        
  • Seizures
